My two biggest timesavers for digi art have been:
merging the sketch and lineart phase. Instead of making a super-rough sketch, lowering the opacity and then doing clean lines on another layer, I now try to keep the sketch relatively clean from the start and then clean it up by erasing and redrawing small sections. This also pushes you to get things right the first time as well as abandon training wheels like eye-height guidelines, which I think is a good thing if you're looking for optimization techniques anyway.
creating color boundaries by wand-tool-selecting the outside of the lineart, expanding the selection by 1-2px and then inverting the selection to grab everything inside the lineart. In the past I'd define color boundaries with the brush tool, painting very carefully beneath the lineart layer, which was very boring and time-consuming.
