Actually it's from Resident Evil. The conversation was between Barry (forgot his last name) and Jill Valentine (one of two playable characters) after they had discovered either the shotgun or grenade launcher, not sure which one it was.

Edit: Infact I found a video of the conversation. Check at 1:45, they don't talk about the backdoor but they do talk about the weapon.

The scene on Gmod Idiot Box Episode 1 at 1:58 is from Silent Hill 2. I remember hearing it while my friend was playing it and recognized it from the first episode. This one, though, that the OP is requesting from episode two, is from one of the old Resident Evils.
