Can you potentially do splitscreen with a multi-monitor setup? Then it'd feel practically the same as playing on two different machines.
Can I use any PC controller, or is it limited? I have one of those PC/PS2 controllers.

While it would be cool, I'm not sure if it would be possible unless you were able to change settings and stretch the game over two screens

No, it will count as two 1080p displays.
Keep in mind that you have loads of depth culling, visibility checks, shadering crap and vertex transforms that might need re-calculation.

I tried to once when my friend was over (because his 360 copy lagged like a bitch) and couldn't get it to work. It may have been functional on release, but I couldn't get it to work when I tried (a few months after release).

Just tried to see if it still works. It still works.
I played Portal 2 co-op with splitscreen on my PC yesterday. Aside from the unproperly configured controls and broken pinging menus it was perfect. I got it to work with mouse\keyboard + PS3 controller, it was a pain in the ass but it was worth it. I couldn't stand playing co-op on the PS3, the game felt like it was running at 20 fps in some areas.
For anyone that wants to know how I did it.
Basically you set up your preferred controller and get it to work and play co-op the same way you would normally for offline co-op on PC just load the game using ss_map "map name goes here". The controller and the keyboard will only control player 1 for now.
The work-around to that requires cheats enabled with sv_cheats 1
You type in_forceuser 1 which will force the keyboard controls to player 2. Then you type cl_mouselook2 1 to force mouse controls to player 2 and you are done. There are a lot of other work-arounds which are very hacky but this is what worked for me and imho its the easiest way with a dualshock 3. The only downside is no achievements.
