Simply put, moments in games that the first time you played them, they made you shed manly tears.
Kicking it off with Red Dead Redemption's ending. I would also post Dutch's final confrontation, but that wasn't so much a manly tears moment as it was just a powerful moment.

Sev being captured/killed/who knows what in the end of Republic Commando.
Even sadder, we'll probably never see a sequel.

Emile's death at the semi-end to Reach. For that matter, any death in any form where the character kind of embraces it with open arms, IE Jazz in the first transformers movie.

Pretty much most of Advent: Rising, seeing your character loose most of his friends in a war he can't win.

All of the people that die in all of the COD campaigns.
I hate it because in COD you always are out with the characters but the majority of them die at the end, whether in be in the game or a sequel.
It's even worse when you check times and each games only spans over about a week.

They always kill off the best characters. Price pretty much comes back magically if he dies anyway so no one cares about him. And as a side note I stopped caring about soap after I saw him with that dumb mohawk.

I believe the Harvest Moon games are kind of sad. I'm playing through the SNES one now. It's not sad as in "omg so-and-so died, i'm gonna cry" but for such a bright kid-friendly game, it has a lot of dark undertones. It's hard to explain, but the game gets depressing if I think about it for too long, but it creeps up on me so subtly that I barely notice it happening.
But in terms of "omg so-and-so died, i'm gonna cry" games, Cave Story was a great one.
