I've read "Detective Comics #27", Batman's first appearance but other than that I have no clue where to start, given that there are so many to choose from.
Any suggestions?

Haven't read much of the new 32 batman other than the Harley Quinn comics, which are pretty good.
Three of the best Batman comics I've ever read, and will spoil you for any you will read in the future are:
Batman: Year One by Frank Miller & David Mazzuccheli,
The Killing Joke by Alan Moore and Brian Bolland,
and
Batman: The Dark Knight Returns by Frank Miller
One not listed so far: Arkham Asylum: A Serious House on Serious Earth
Otherwise, Year One and The Killing Joke are must reads

It does modify the story a bit and Batman's monologue is gone though, also has some pointless line changes. But yes, is definitely the best DC animated movie.
