I'd assume 1 person on mouse and keyboard, the other on gamepad.
Does it support two gamepads? It was intended for Big Picture after all.

Yeah, unless they fixed it, I could not get splitscreen to work with any combination of keyboard+mouse/controller players. It only seems to support two controllers.
I'm sure some modification could easily enable mouse+keyboard for one player and controller for the other; that's how I used to play Left 4 Dead.
I was able to activate splitscreen with 2 joysticks, but I could still control my character with M+KB while my friend could use the 2nd joystick.
Couldn't get it working using just mouse+keyboard and controller, so I think it's controllers only
