He was reportedly found dead while on holiday with other players at the Flamingo Hotel.
Flamingo Hotel spokeswoman Debbie Munch confirmed to ninemsn a person had died at the hotel.
"A gentleman was found unresponsive at 5.40am local time on the southern driveway of the Flamingo Hotel and Casino," Ms Munch said.
"He was transferred to the University Medical Centre of Las Vegas where he was pronounced deceased."
It has been reported that the player fell from a balcony at the Flamingo Hotel.
Ms Munch could not confirm details of the man's death but said the Clark County coroner's office was investigating.
The football club released a statement today saying it was in contact with members of the travelling group and was working with US authorities while investigations continued.
"At this point, the club is unable to confirm the identity of the player involved in this tragedy as family members are still being informed," the statement reads.
Port Adelaide general manager for football Peter Rohde is on his way to the United States to be with the players.
The club has also made contact with its other players still in Australia.
A spokesperson for the Department of Foreign Affairs and Trade said the department was aware of reports an Australian man had been involved in a serious incident in Las Vegas.

Pretty sure they have confirmed it.
They think he fell off a balcony and they found him on a drive way.
