A man has been arrested and charged with the theft of the outback ambulance that nurse Gayle Woodford was seen driving before her disappearance on Wednesday night.
Police say they are throwing all available resources into the investigation.
A wide search is being made of the remote APY Lands around Fregon and Marla and police say they have serious concerns for the safety of the 56-year-old woman.
Police said an ambulance fitted with a GPS left Ms Woodford's home on Wednesday evening and was driven from Fregon to Mimili in the early hours of Thursday.
They believe Ms Woodford was at the wheel of the ambulance when it left Fregon.
The GPS showed it arrived in Marla about daybreak after travelling via Indulkana, police said.
When the ambulance got to Coober Pedy mid-morning on Thursday it was intercepted by police.
The nurse was not in the ambulance but two men and a woman in the vehicle were interviewed by police and one of the men arrested and charged with theft of a motor vehicle and driving while unlicensed.
The Fregon man, 36, has been remanded in custody.

The prospect doesn't sound very good. Either she's been murdered or she's died from exhaustion in one of Australia's most inhospitable regions.
Yeah, It's like unlikely she'll be found given that area is pretty damn hard to survive in. She may get lucky and find water or tribes in the area.
