Fuck bitches that cheat, it'll be pretty ballsy, but you should tell him. It's always better to be honest in a relationship, even if you want your sister to find love/happiness/etc. you should tell him, mostly because if she can't be honest with any of them then she won't be a good girlfriend regardless, you'd be saving a lot of grief down the road if you help end it now.
But be prepared for a lot of anger, and probably some yelling, and some hatred too, that may come up a bit. But if you're any sort of man and want to help your sister and the guy you should tell him.
