Before people say 'Uhg, another ideas guy,' I can help map for this gamemode, despite me not being the most amazing mapper in the world.
Anyways, onto the gamemode. Have you ever played a DoTA style game, like LoL or Monday Night Combat? Coming Towers, Character Skills, and NPC attacks, you get DoTA style. Well, why not make a gamemode like that for Garrysmod? It would be some work, sure, but I bet it'll make a nice end result.
There would be 2(more?) teams, most probably Combine and Rebels. Small groups of Rebels and Combine sp0awn at intervals, and head towards enemy bases. From the base, in specific spots, players can build/upgrade 'towers,' or 'sentries,' or 'turrets.' Perhaps even select areas where you could build some sort of 'harvester' towers, which slowly gain the entire team money, energy, or whatever. Some custom mobs would need to be made, like a Boss Mob that spawns ever now and then for each team. The rewards are great from these, as they have astounding health and very large damage splashes. For the combine, probably Striders or APCs (if possible), but I'm not sure what the rebels would get.
The order of spawning would go pretty simple. Maybe one or two stronger NPCs, and the rest very weak. Every now and again, like I said, bosses. And maybe allow the players to purchase upgrades for their NPCs, and possibly allow them to buy one wave (spawned only once, unless bought again) of special units, the better the more expensive.
Well, thats just my idea. Hate it, love it, or 'meh' it.
You never know. Besides, the NPC spawning could be created just directly in the map on its own, without lua scripting. But turrets, classes, custom NPCs, and the rest, prolly needs scripting.
I don't have a clue what DoTa is.

DoTA, is basically a mix of Tower Defence and a few other things. You build 'towers', probably turrets in this case, while friendly mobs spawn at intervals at your base and attack enemy bots and players all the while heading to the enemy base in attempt to destroy it. Players also help destroy it, aid their bots, build 'towers,' and kill eachother.

Oh yes, and the towers can only be build at predetermined locations, unless there's a class that can build mini-towers where he wants or something... look at Monday Night Combat at the Steam Store, its a DoTA-like game.

This has been made multiple times and it has always been private but as Im soon starting my community containing 4 custom gamemodes I am looking for some new shit.
I will probably give this a try seen as the servers I have played on I loved this so much.

Its not just tower defense. In fact, tower defense is the least crucial part. NPC spawning, base protection, and killing other players is.

In other words-- the towers are there as a last resort, if the enemy players and NPCs break through your teams NPCs and your players.

Why not? A lot more complicated things have been done.
I had an idea to make a dota gamemode before the only thing stopping me from doing it is the click and moveto player movement, I don't want to do it hackly and use a NPC I want to move the actual player. Last time I tried to do it I failed but I may try again and if I get that working I will take a shot at it.

No its nothing like that
It works this way:
The two sides have their bases.
The structure consists of:
3 Lanes. (Paths)
3 Towers per lane.
Enemy Respawner.
Enemy Enforcers (kill em', the enemies of the opponent gets weaker/slower/less respawn)
The respawn (Can't destroy this one)
The Creeps respawn (Cant' even see this one)
The Creeps (Little monsters that'll give you EXP/Gold in the beginning, and will also help you rush the enemy base)
The Main Base (A.K.A. Nexus, Etc.)
Some other random stuff I forgot.
Also, each player controls a "hero", there are 2 teams, If my memory is not playing tricks on me, you can do 1x1 ~~ 4x4.
There are levels and items, items are made with recipies, that consist of the recipe scroll and the sub-items.
There are also the skills, that you can add up once you get skill points, wich happens every time you level.
The objective is to destroy the main building (nexus/etc.)
TO understand it better, play HON / DotA or LoL (League of Legends).
