There's already a game based on Rock Paper Scissors with a twist.

It's called Pokemon.

I remember when I was little I took a deck of around 42 index cards and made my own simple rock paper scissor game.
It would have something like "Virus" which would kill all organic things, but "Anti-virus" or "Hazmat-suit-guy-man-thing" could beat it.
It had other things like dog beats cat, water beats fire, nuclear-warhead beats everything.
It was a neat little game I invented.
