I'd recommend waiting until at least level 32 - ideally the level you're hitting Sanctuary in NG+. Damage ramps up significantly between the modes, and a good weapon in the low 30s can make your life a lot easier. Really, though, you can throw all caution out the window if you have multiple keys. You have a pretty good chance of getting something good for your level regardless of when you spend it.

I used my key when I was lvl 5.
