I'm guessing they might be something that builds structures, kind of like humanoid ants. That'd be pretty cool, if you think about it- you're wandering along and HOLY SHIT A CASTLE. I don't know, just explains the block holding.

enderman's eyes should stay bright white in the dark, as spiders have with their eyes.
They should toy with you. Like you wake up and walk outside and there's a deep trench dug around your house. Or they leave you a chest filled with string.

They should get really close to you and interfere with you until you're basically forced to look at them. Like complete assholes.
That won't be in the mechanics, but this will still happen to me, no doubt.

I think Enderman's eyes should be red when they are attacking you. Imagine this:
You are outside mining away some dirt for a dirt fortress. After all it's easier to get then stone so why not? The sun begins to set above you. You think nothing of it I mean you have an diamond sword, and a bow. So what could hurt you. Soon enough you begin to turn around to your house when you spot one. Although you already know about these Endermen from the forums so you don't look directly at it, but suddenly you hear a "SSSSSSS" behind you! Flipping around with your sword at hand you cut the creeper in half right there. A terrifying scream fills the valley you are in. Spinning around you just see 2 huge red glowing eyes. Then nothing. You wake up on a beach with no items. You died.
(If you don't get it when he turned around to kill the creeper his curser pointed right at the Enderman for a split second.)

I think teleporting is just another cheap trick to avoid having to code pathfinding. Dogs already teleport, and while you're looking right at them (they still manage to kill themselves a whole lot). The creepiness could be so much better if the endermen and all other mobs could be found trying to get in through doors and things that you actually built as access points rather than walking up to walls and humping them, falling off cliffs, or just appearing behind you (hurr durr).
I think it'd be more creepy if they didn't interact with you, like you just see them watching you sometimes.

But how would you react if you saw a blank face looking at you from behind a hill? It gives you an eerie feeling. Especially if you didn't already know they're harmless.

Or something like this in the video. If you have played Ocarina of Time then you might understand where I am going with his.
When you explore a cave or maybe a ruins there is a small area that spawns paintings of the Endermen and when you destroy the painting maybe 1-3 Endermen are spawned and you kill all of them that are spawned. And when you destroy all of the paintings then you gain a reward depending how many Endermen you have killed or paintings you destroyed.
Maybe gold or a rare weapon or item? this video is kinda the base idea for it:
