After the trailer, an announcement by Rockstar said it was in Los Santos and the surrounding hills, countryside, etc. However, with GTA: SA they said a similar thing, but ended up with 3 cities, and also for GTAV, people from Rockstar North went to the real-life equivalents of all 3 cities in San Andreas. However, don't get your hopes up that it's all of San Andreas.
well it clearly is Los Santos, I noticed it in the trailer now that I watched it again (beach gym shows it, and there's the vinewood sign there too) I'm hoping for more open spaces. No need for a desert like in Las Venturas, but just some more space than IV had. I felt that there wasn't much to do there.

[editline]3rd December 2011[/editline] also noticed a fair bit of attention to detail, the clouds above the mountain are moving, and wind affects trees and shit now. pretty subtle but cool [editline]3rd December 2011[/editline] also just noticed what looks like a water plane above the uhm windmills probably late with all of this shit but in the time I last saw people pointing stuff out I didn't hear about it so

oh god wingsuits PLEASE WINGSUITS [editline]3rd December 2011[/editline] oh also that house with the for sale sign has an open window with it's curtains flapping dunno if that's been noticed or what [editline]3rd December 2011[/editline] only other thing I can notice that seems different (though could be argued is scripted for the trailer) is the NPC interaction, seems much more natural like they actually have behaviours rather than just walking around saying random lines and interacting with different npc groups through simple love/hate relationship. I mean, yeah in gta iv you'd see people having conversations and shit, but people checking people out, getting thrown out of bars and stuff at night... it'd definitely make the world feel more living... what if they have added some kind of super dynamic npc interaction? i mean they've already got euphoria licensed which has pre-set behaviours for ragdoll and stuff, but euphoria can also be used for other types of realtime behaviours, it's not just for ragdolls... would be interesting if they did anyway

If anything, they keep every physics (and in general, every improvement) they make. But this largely goes for the environment. Because science always moves the same way! But with things like character movement and (gun)/fighting and shit like that, that will always be adjusted to fit the theme of the game.

K-ROSE was the best station in SA.
