Does that mean you can fight against him but not play as him?
or it means there are more characters to play?

Rain is pretty much confirmed unplayable. What you saw was likely story mode, since he does appear there. Maybe Tower gameplay has him at times too.

He is a NPC (not playable character), this means you can play against him but not use him, thats why he has no variation..... same with a bunch others chars in the story
However, Tanya is also a NPC in the story, maybe he will likely have the possibility of being a DLC in the future

It seems that they've put a filter on his voice to have a bubbly underwater feel, but it colliding with the voiceactor makes him sound like a cross between Sean Connery and DKR Bane.
This is the best possible outcome.

She is; the NPC Tanya borrows from other characters. When Tanya releases, she'll have her own animations, moveset, and variations.
