I tried out the Daily Run but had to quit instantly because I noticed I had leftovers from a custom run I made with run editor (Leviathan with Maw of the Void, Abaddon, and Pentagram with a bunch of damage nerfs). Is making an illegitimate run seriously that easy? Is there nothing keeping me from just booting up Ultimate nuclear Tammy homing Brimstone against the run and getting a ridiculous end score?

This run has some pretty powerful items, blue candle, red candle on a later floor, keepers pockets when you don't need that item I think.
BASEMENT seed is pretty silly with the dark room teleport. It doesn't loop in the dark room, but those runs technically have an end other than death now.

In the same shop there's a Chaos Card just waiting for you.
Today's daily lets you get car battery + blank card + heirophant, allowing an easy sacrifice room skip to the dark room. The target is cathedral though, so your time is attributed to 6th level and not the 10th.

Cain's Other Eye now scales with your damage
Shoop Da Whoop now deals x4 tear damage instead of x2

New bug:
Finishing a run as Judas's Shadow (and presumably Blue Baby from Ankh and Lazarus 2) unlocks the item and achievement for your original character, but the completion mark still goes to the character you transformed into
sacrifice rooms can warp you to dark room during a BASEMENT seed run

Okay other things I missed from earlier.
Flagellation in a sacrifice room [i]overrides[/i] having set a precedent to get Devil deals. However it's not 100%.
Shoop da Whoop damage doubled.

Picking up the Match Stick will force the Tick to be dropped.
Afterbirth fly-based items count toward Beezlebub transformation
Cain's Other Eye scales with your damage
Friend Zone now deals ticking damage at a proper rate instead of like once a second.
Taurus now recharges on a per-wave basis in Boss Rush and Greed Mode instead of being useless
Quit/Continue no longer gives you an instant Boss Rush clear.
Mac crashes should be fixed now
Visiting Mega Satan before unlocking the Angels is no longer possible. The door just doesn't spawn until you meet that requirement.
Finishing a run after transforming into another character now grants rewards for the original character, but marks completion for the character you transformed into. One of those things shouldn't be happening, but it undermines the Missing Poster's value for now since you wouldn't unlock any of Lost's items unless you started as the Lost.
Butter Bean got buffed, what a day to be alive. Since it uses the same code, this also means that Farting Baby, Mysterious Candy, and Butt Penny got a buff as well.

If you ever wanted to check out a specific player's leaderboard rankings in your browser instead of in-game and needing to be their friend, you can now. It parses through scores to flag potential cheaters and prune them from the rankings as well. The source code is public.

I think it's safe to say at this point that it was meant to be lol, but for whatever reason they ending up releasing what they had done at the time, then patched in more content as it was ready. Not sure why they're now packaging the rest as DLC, especially after that sacrifice rework that Edmund originally said was cut entirely, but whatever I'm just happy we're getting it since this game is my crack.
I do hope it expands the blue womb some more but if not hey it's still more content.

No more than 5 minutes for both basement floors, and I'd say the other 15 is for the rest.
Prioritize gold rooms and shops, and then go straight to bosses.

It also depends on the items you get, if you get something like Mom's Knife early on then Boss Rush is guaranteed most of the time.

Do your business and keep moving. Don't bother with arcades unless you're there to blow shit up for the rewards or to knock your health down so that you can check a boss gauntlet room to see if it has something worth fighting for. If you don't find a secret room on your first pass of the floor, don't bother looking. You're going to want either the Treasure Map, Compass, or Blue Map if you can swing them as any of them on their own can indicate where the boss room is likely to be. The only rooms of importance to you should be the Treasure Room, Boss Room, and maybe the shop. Make sure you can consistently get Devil Deals as the items offered will typically give you a good boost in how quickly you clear rooms and keep moving. If you're playing as Isaac, try to keep rerolling to a minimum as the time spent backtracking won't do you any favors; just take your shit and keep walking. You typically won't need more keys/bombs than what's given to you, so farming is typically unnecessary. Try to avoid spending them on frivolous things as your focus should still be on those 4 item-yielding rooms.
So long as you stay aggressive and resist the urge to min-max, you can pretty reliably make it to either Boss Rush or Hush, sometimes both if you're strong enough.

Don't overlook the value of the Map item or the Compass item from a shop and save up on bombs if you think you're going too slow. In case of emergency, you can use the compass or even just a blank map to guess where the boss is (small room furthest away from you), then use bombs to skip all the rooms and quickly get there. If you get an anzus rune, save it for depths 2 so you can do the mad dash.
Never collect Proptosis+Polyphemus+Rubber Cement.
Your tears are huge and bounce off everywhere instead of hitting the enemy.
It nearly ruined my run and was really annoying.

I basically go for nothing but the treasure room and the boss room. Ignore everything else if possible. MAYBE a shop if I have 2 or 3 keys and over 15 cents.
Other rooms like arenas and arcades might be helpful, but they take too much of a time investment to be worthwhile if you want a boss rush. Same with shooting fires and poops and all that. The reward is too little for all the time it'll take you.

if it was free I feel like they wouldn't be calling it a DLC, they'd just keep patching it in like they did with the sacrifice room. it's gonna cost $$ I'm sure, just probably a lot less than afterbirth.
