Hour by hour maybe 15-20 depending on how fast you read, however you're unlikely to read it all in one sitting.

It took me three days to get from the start until I caught up (Jade: Enter), but that was literally all I did for the three days. If you casually read it, then Two Weeks sounds like a good estimate.
Don't skip any parts or intermissions though, they all have details of the story you won't want to miss.

It took me about 2 weeks of very casual reading to catch up, and thats including getting bored and leaving it for a few days during act 5.

seriously the worst thing you can do to yourself while reading homestuck is allow fan content to influence your perception of characters

I started reading somewhere around September 2010, didn't take me that long. (A few days, I think.) Of course, it was still Act 5 Act 1 back then.
Took my little sister something like 3 months. She took her time though.

if I recall correctly it took me around two weeks to finish homestuck due to school and being generally discouraged with reading it once I reached the part where the trolls showed up. I hated the trolls on my first read through.

For you guys who were caught up before Act 5, how long did it take you guys to get impatient (Assuming you did at all) with them?

i didn't really like the trolls until act 5 act 1 was over because i thought they weren't going to be important
then it was revealed they created the kids' universe than they became more likable
plus the convos with them and the kids were really funny a lot of the time
I didn't like them because they weren't the Midnight Crew or Dave.
