Sounds like he didn't quite get WHY people gave him that advice, because of course you can tell a joke in any different number of panels. But much every simple story can be split into "beginning, middle, end", many jokes also have a beginning where you familiarize the audience with the setting, a middle where you set up the pun, and finally the punchline at the end.
Many jokes benefit from the three panels format because it can embody the above at the fastest pace possible if you reserve one panel for every part. It helps people getting their pacing straight (whether they usually take too long for a joke or forget telling the audience about crucial details) and pacing is crucial for many jokes. That's why it's helpful.

But he didn't seem to get that and thus thought it would be nonsensical to blindly do whatever people tell you to do.
