PNG for few colours. (vector graphics, aliased ms paint comics and simple shit)
I always compare the two whenever I want to save something though, chosing whichever has the smallest size.

What?
PNG has superior quality to JPEG, and you should only save as JPEG if your PNG file would be huge. If it's less than 1000 pixels in both dimensions, then it's probably safe to save as PNG.

No. If you save with JPEG at quality 12 (maximum) then it's as good as loseless in visible quality. There is NO VISIBLE difference. The difference will only be noticed when you edit the image again, but you can save it over and over as much as you want in quality 12 and there will still be absolutely no visible difference, it will look like the original. It's only the information in the picture that gets changed, something that will only be visible when you hold in ALT whilst dragging the levels slider.
People have just been completely misslead by MS Paint jpeg (which can't change the quality) and people boosting about how "superior" PNG is even on files it shouldn't be used on. I remember a year or two ago here on the forum someone boasted about PNG being better in a thread where a guy had really visible JPEG artifacts on his picture, so being convinced he took the JPEG compressed image in again and saved it as PNG and he said "Yeah, I can see that quality is a bit better". I think that serves as a good example of how easily people are fooled. Goddamn placebo effects.
JPEG and PNG are end-files, they aren't meant to be edited again, so only the visible result matters.
I'm also a child burnt by MS Paint, so if I want to show somebody something I usually pick PNG, and when I'm worried about file size I save it as JPG with a quality of 9 or something, which is usually a good compromise.
This way I usually don't have to use the JPG quality slider (oh the back-breaking additional work) and I don't accidentally save images with transparency as JPGs.

Only if you don't plan to make it anti-aliased only for the sake of using the bucket tool almost constantly with only adding little to no shading.
