Nope. They started in the 2530s, I think, on Onyx. The events of Ghosts of Onyx happen concurrently with Halo 2(well, technically after Tartarus activates Halo Delta, so it goes a few days after Halo 2's end), so the third class of SIIIs were in deployment by the time Reach fell.

The SIII's were injected with certain illegal compounds during the augmentation
process that caused them to rely more on their primitive, "lizard mind" during priods of extreme exertion. The side effects of said compounds could also cause mental degeneration in the SIII's. Which would make them extremely violent, and insane. To counteract these side effects, the SIII's had to have regular injections of other chemicals that would keep them from going bat shit insane.
tl;dr version:
Spartan III's were injected with stuff.They have to be injected with more stuff every so often so they don't go crazy.
