Revengeance is relatively standalone. It has a couple of off hand references to the main series, but not playing them isn't going to ruin MGR for you at all. Though it would probably help if you want to actually know who some of the returning characters are.

It is standalone but also a sequel to MGS4

They actually don't explain anyones powers other then Vamps. People like to say that they explain Ocelot getting taken over by Liquid as therapy but they forget that Ocelot has a completely different arm in MGS4 then he does in MGS2 which is why he underwent the therapy because he noticed that he seemed more unpredictable when Liquid was taking over
(there's also the whole Ocelot's Jap VA died so Kojima had to jump through some hoops)
