"Cis" is the opposite of "trans" - it comes from Latin, where "cis" means "on the same side as", while "trans" means "across from". You see it a lot in organic chemistry, where many isomeric compounds general have a "cis" version, and a "trans" version where half of it is flipped.
A cisgender is the opposite of a transgender. Transgenders are "a <gender> trapped in a <other gender's> body", while cisgenders are "a <gender> trapped in a <gender> body". So if you're a guy who thinks he's a guy, or a girl who thinks she's a girl, you're a cisgender. That doesn't have anything to do with sexual orientation - you can be a gay cisgender, or a lesbian transgender, or all those things.
The joke, in any case, seems to be either a parody of what conservatives think would happen if liberals ran everything, or a parody of Tumblr and the like where people care way, way too much about "____ privilege", and often have odd ideas of "equality". They would also take heavy offense to what I said in the second paragraph, since I completely failed to go into the difference between sex and gender or talk about gender roles or any of that bullshit that really doesn't matter in the grand scheme of things.
