As long as the singleplayer is the main focus, simply modifying the maps slightly for co-op and having a sort of "Who can assassinate the target first, and the most stylishly" and "Team up to assassinate" would be very interesting.
Can imagine it starting with you already having a civilian disguise which the other guy can't distinguish, so you not only have to track your target, but watch out for the other assassin.
Or perhaps a 1v7 thing, where 7 players are elite bodyguards who must find and kill the hitman before he can assassinate the target.
Pretty much these play out like AC:Brotherhood, but with guns and hitman style.
That said, The only version I think would really work well would be co-op. Co-operative assassinations could be deeply satisfying, as my experiences playing splinter cell: Chaos Theory co-operatively can attest to. Co-op stealth works really well.

Imagine it: One of you works on getting up to the roof, whilst the other infiltrates their way in, getting close to the heavily guarded target; no way you could just go up and shoot him from there... but you impersonate the guy you're dealing with, suggest you go in a nice, wide, open space to do the deal; no chance of an ambush.
When you're there, you exchange the money and, just as you take the briefcase with the untraceable blood diamonds, *TAKKA* *TAKKA* from the semi-automatic sniper rifle, and as the next two turn to where the shots came from *fwtup* *fwtup* from your silenced silverballer. 1 dead target, and 3 bodyguards. The up-close assassin hides the bodies in a nearby bush, and uses the bodyguard's clothes in order to negotiate a way for the sniper hitman to get off the roof, making sure guards are elsewhere, distracting them and so forth.[/QUOTE]
I agree. Coop assassinations could open up some of the most interesting options yet for hitman.
If nothing else it would make the last two games they have made useful for hammering out netcode of cooperative games.
Please no multiplayer.

Kane and Lynch was a doomed prospect from the start, IO should have just stuck to what they were good at (Hitman).
The ARG is over! We now have information that something Hitman-related (most likely Hitman V) will be announced at the first day of E3!

Remember in the mission at the rehab center? Same stuff used on the agent in that.
I hope use new technology to their advantage, things like Euphoria and so on.
I'm rarely pessimistic but after Kane and Lynch 2 Troll Days I have a feeling this is only going one way and that is down.
Which is a shame because every Hitman got better with each instalment. If they add a cover system and make it more "ACTIONY BADASS MERICA FUCK YEAR" I will be disappoint.

That would be great, but i hope it isn't used as much as in Gta 4, i dont want to be knocking people to the floor just by walking into them, it'd be amazing if bodies actually had some weight to them though, and the physics system wasn't as terrible.
