I know for sharks you need microscope slides of their spinal cord to get an accurate measure of their age, organ contents might also require lethal research, stomach samples, tissue samples, etc. However, Japan has had a hard time proving to the IWC that there are no non-lethal methods to get the same data, and their quotas are so large that it's quite obviously a cover for commercial whaling. Not to mention the Southern Ocean hunt is not their only hunt. They also have Pacific Ocean hunts and annual coastal hunts.
