You have a change in the DNA of a few cells that breaks some regulatory process, either for growth control or DNA copy correctness. If the wrong regulatory process breaks, you end up with rapid growth in this independent rogue line of cells, potentially with each successive generation more broken than the last.

He does have cancer, but he's not just going to die anytime soon. I don't know how realistic it is, but he is optimistic about an eventual full recovery.
